Overview

Primary human CD4+CD25+CD127lowFOXP3+ T cells are isolated from peripheral blood mononuclear cells (PBMCs) using postive immunomagnetic separation techniques to isolate particle-free CD25+ cells, and unwanted cells are subsequently targeted for immunomagnetic depletion. These cells are often referred to as regulatory T cells (T regs) and are used in immune response assays.

Cells are obtained using Institutional Review Board (IRB)-approved consent forms and protocols, and cryopreserved in animal component-free CryoStor® CS10 medium.

Figure 1. Cryopreserved Tregs Suppress CD8+ T Cell Proliferation

Human Peripheral Blood Mononuclear Cells (PBMCs), Frozen (Catalog #70025) and PB CD4+CD25+CD127lowFoxP3+ T cells (Tregs), Frozen (Catalog #200-0120) were co-cultured for 4 days at various ratios of Treg:PBMCs in the presence of 2.5 μL/mL ImmunoCultTM Human CD3/CD28 T Cell Activator (Catalog #10971) and ImmunoCult™-XF T Cell Expansion Medium (Catalog #10981). The Treg-mediated suppression of CD8+ T cell proliferation ranged from >90% at Treg:PBMC ratios of 1:1 to <5% at Treg:PBMC ratios of 1:32. Experiments were performed with 3 independent Treg donors, represented by the gray, orange, and black lines (mean ± SD, n = 3).
