Overview

Use CellPore™ Delivery Cartridges with the CellPore™ Transfection System to enable cytosolic delivery of diverse cargo, including nucleic acids (e.g. siRNA, mRNA, self-amplifying RNA), ribonucleoproteins (e.g. Cas9 RNPs), proteins, and other biomolecules to a variety of cell types. These cartridges feature integrated microchannels (constrictions) of specific dimensions that leverage mechanical deformation to achieve transient non-toxic disruption (mechanoporation) of the cell membrane. This physical delivery method offers a gentler approach, making it an ideal solution for sensitive and hard-to-transfect cells while preserving cell viability and function. Each cartridge comes pre-assembled, and includes an insert for handling the cell & cargo mixture, as well as its collection tube for easy and streamlined post-mechanoporation handling. Together, each CellPore™ Delivery Cartridge supports sample volumes ranging from 20 to 200 μL.

CellPore™ Delivery Cartridges are available in five different designs to ensure optimal mechanoporation for a variety of cell types. The numerical designation corresponds to the constriction size, with smaller numbers designed for mechanoporation of smaller cells and larger numbers for larger cell types. This range of CellPore™ Delivery Cartridges supports high-efficiency delivery across a broad range of cell types, including primary human and mouse immune cells, stem cells, and immortalized cell lines. Reference our CellPore™ Mechanoporation Guide to determine which cartridge is best suited for your cell type or application.

CellPore™ Delivery Cartridges are recommended for use with CellPore™ Delivery Media and the CellPore™ Transfection System to support optimal transfection efficiency and viability during mechanoporation.

CellPore™ Enables Efficient Delivery of Cargo to Primary Immune Cells, Stem Cells, and Cell Lines

Figure 1. CellPore™ Enables Efficient Delivery of Cargo to Primary Immune Cells, Stem Cells, and Cell Lines

Cells were resuspended in appropriate CellPore™ Delivery Media and transfected using the CellPore™ Transfection System. (A) Cas9 RNPs targeting TRAC (TCRɑβ), B2M (MHC-I), CIITA (MHC-II), TIGIT (TIGIT), KLRC1 (NKG2A), or PTPRC (CD45) were successfully delivered to various primary immune cells, stem cells, and immortalized cell lines, resulting in the knockout of their respective surface marker with minimal impact to cell viability (relative to untreated controls). (B) Representative flow plot of co-delivery of mCherry and eGFP mRNA to human unactivated T cells, which resulted in over 90% co-expression of both constructs. (C) Delivery of a self-amplifying RNA construct encoding GFP (1.5 pmol, 5 μg) to human CD34+ hematopoietic stem & progenitor cells resulted in successful GFP expression expansion over 7 days (with 0.8 μg/mL puromycin; n = 2). (D) Delivery of a 3.5 Kb GFP-encoding plasmid DNA to Jurkats resulted in dose-dependent increase in GFP expression and maintenance of high viability (n = 4-6). Results measured via flow cytometry. Untreated refers to unmanipulated cells. No Cargo control refers to cells mechanoporated without cargo. Data presented as mean ± SD.

CellPore™ Mechanoporation Performance in Human Unactivated T cells

Figure 2. CellPore™ Mechanoporation Performance in Human Unactivated T cells

All experiments were performed using CellPore™ Delivery Cartridge 300 and CellPore™ Delivery Medium A with 2x106 cells per 50 μL reaction. (A) FITC-Dextran (0.1 mg/mL) was delivered across a range of pressures, and optimal intracellular cargo delivery and cell viability were measured between 70 - 90 psi (n = 7). (B) Independent or co-delivery of eGFP and mCherry mRNA (15 pmol, each) at 90 psi resulted in high expression efficiency (> 90%) across T cell subsets, measured by flow cytometry 24 hours post-mechanoporation (n = 3). (C) Cas9 RNPs (60 pmol; 1:2.5 Cas9:gRNA) targeting B2M (MHC-I) or TRAC (TCRɑβ) were delivered at 90 psi, resulting in > 80% surface marker knockout efficiency while retaining high cell viability (n = 5). “Untreated” refers to unmanipulated cells. Endocytosis control represents natural uptake of FITC-Dextran in unmanipulated cells. Results were measured by flow cytometry. Data presented as mean ± SD.

(C) Fresh NK cells isolated from peripheral blood were cultured in ImmunoCult™ NK Base Medium supplemented with 10 ng/mL IL-2 overnight prior to Cas9 RNP delivery (60 pmol, 1:2.5 Cas9:gRNA).
